A completely reshaped front row sees Gregor Hiddleston and Zander Fagerson promoted to the starting lineup after featuring off the bench in Johannesburg, while Patrick Schickerling returns at loosehead following a rest during the opening match of the South African double-header.
Burke earns his first senior start just a week after debuting for the Warriors, partnering Alex Samuel in the second row.
At openside, Angus Fraser starts in the same week he agreed a contract extension, joining Euan Ferrie and Jack Dempsey.
In the backs, Ben Afshar is at scrum-half, while Adam Hastings returns from a head injury sustained against RC Toulon to start at fly-half.
Midfield continuity sees Huw Jones remain at outside centre, with Sione Tuipulotu coming in at inside centre after his bench appearance last time out.
On the wings, Kyle Steyn returns to captain the side, while Kyle Rowe shifts to full-back to allow Ollie Smith to start on the wing.
Among the replacements, Johnny Matthews and Sam Talakai return after missing last weekend, with Nathan McBeth completing the front-row cover.
Dylan Cockburn keeps his place on the bench for a second straight match, while Jare Oguntibeju and Sione Vailanu rotate to the replacements after starting at Ellis Park.
The bench is completed by Jack Oliver and Stafford McDowall, the latter having scored in each of his last three appearances for the club.
