Scott Cummings, Zander Fagerson, Huw Jones and Sione Tuipulotu are among eight Scots included in Andy Farrell's 38-strong squad.
Bath's Finn Russell, Blair Kinghorn of Toulouse and Edinburgh's Duhan van der Merwe and Pierre Schoeman are also included.
For Tuipulotu, it means a return to the land of his birth but his place in the squad was was put in doubt by a pectoral injury that ruled him out of Scotland's Six Nations campaign.
The 28-year-old centre expects to feature for Warriors over the next couple of weeks and is grateful for the faith Andy Farrell and his Lions coaches have placed in him.
"I just want to take my opportunity now," Tuipulotu told BBC Scotland. "I just want to repay the coaches by jumping back in there and proving why they selected me.
"I feel like I'm going to be a better player than what I was when I got hurt. I'm going to be more hungry, more desperate. I'm so excited.
"I'm not embarrassed to say that since the year started it's been all I could think about... playing well for Glasgow, for Scotland, so I could play for the Lions."
Tuipulotu has not played since that training injury but it now close to a return.
"My dad was over at that time and he'll be able to tell you I was walking around the house going, 'I guess that's me'," he said. "I just thought that was the way that it panned out.
"I was playing the best rugby in my life and I got hurt. You battle your own emotions and stuff like that all the way up into selection saying, 'am I even still being considered?' and stuff like that.
"But then it all, the culmination of when finally your name gets read out, I can't tell you that feeling. It's euphoria, it feels like.
"I just want to go out in Australia and perform in big games for such an amazing team. The Lions is rich in history and I just want to add my piece to that book."
Congratulating his players following selection, Glasgow Warriors Head Coach Franco Smith told glasgowwarriors.org: "We are all extremely proud of our boys for being selected for the 2025 British & Irish Lions tour of Australia.
"Scott, Zander, Huw and Sione have each contributed significantly for club and country over recent seasons, and the reaction of everyone in the room when the announcement was made spoke volumes about the culture within our environment.
"Their selection is also a reflection of the hard work put in by everyone here at Glasgow Warriors; our medical and strength and conditioning teams have worked with each of the quartet this season to put them in prime physical condition, and our coaching group have worked closely with them to help them be the best versions of themselves.
"Our Lions are superb examples to our young players here at Scotstoun and to young players across Scotland, and we look forward to cheering them on as they tour Australia in the famous red jersey."
Managing Director, Al Kellock, added: "On behalf of everyone at Glasgow Warriors, I want to offer my heartfelt congratulations to the four players selected for the 2025 British & Irish Lions squad.
"This is a moment of immense pride not just for the individuals involved, but for the entire club and our supporters. To have four Warriors named - the most we've ever had in a single Lions tour - is a historic achievement and a testament to the hard work, consistency, and character shown by these players.
"The selection highlights the strength and depth of our squad and the positive environment we've built at Scotstoun. It's a clear reflection of the standards we hold ourselves to and the direction we're heading as a club. We're all incredibly proud and will be backing our lads every step of the way."
