Scotland will compete in the inaugural Quadrangular tournament with their first match in Durban, against Samoa on June 8 and their second a meeting with the Springboks in Nelspruit a week later.
The Scots will bring down the curtain on the trip with a final test on June 22 in Pretoria with the opposition depending on the results of the first two weeks' games.
After a successful season on the domestic front, Glasgow's Tim Swinson, Pat MacArthur, Alex Dunbar, Peter Horne, Peter Murchie and Tommy Seymour could all play their first games in a Scotland jersey.
Matt, the Glasgow and Scotland defence coach, told the Evening Times: "For all these guys this tour will be a great finishing school for them in terms of the experiences they pick up.
"To be lining up against South Africa at Nelspruit, which is a spiritual homeland of South African rugby, is one of the great tests of an international player.
"The Glasgow lads are coming off a great season and are hungry for this challenge.
"I have no doubt all the lads will come back from this tour with experiences that will only make them better players."
